Mitochondrial respiration analysis of T cells exposed to chemotherapy (short-term analysis). (A) OCR analysis of T cells exposed to chemotherapy and either left unstimulated or stimulated with CD3/28 beads for 48 hours. Deficits in OCR are revealed in this short-term stimulation assay. (B) ECAR analysis from the same experiments. Only cytarabine (Cyta) lowers ECAR in stimulated cells. (C) SRC in the same experiments. This measure of mitochondrial reserve is lowered by higher doses of cyclophosphamide and cytarabine in unstimulated cells but these effects are largely gone after short-term stimulation. (D) Conversion of glucose to lactate after chemotherapy exposure and short-term stimulation. Chemotherapy reduces the capacity of T cells to produce lactate.
