Multiple myeloma (MM) cell-derived Jagged promotes angiogenesis: (A) Up: Jagged1 and 2 knock-down (KD) efficiency in RPMI8226KD and OPM2KD cells was obtained by qRT-PCR assay of the relative gene expression variation (normalized to GAPDH) of Jagged1 and Jagged2 and the Notch target gene HES6, calculated by the 2−ΔΔCt formula. Data are expressed as the mean value ± SD. Down: Western blot analysis of Jagged1 and Jagged2 in HMCLsSCR or HCMLsKD. Protein loading was normalized to -actin. The shown images are representative of three independent experiments. (B) Up: Tube formation assay on Matrigel with co-culture systems of HMCLsSCR or HCMLsKD and primary human pulmonary artery endothelial cells (HPAECs). 4X magnification images are shown. Down: Graphs show quantification of the number of areas and branch points and total tube length. (C) HPAEC tube formation stimulated by Jagged1 and Jagged2 peptides. Images are a 4X magnification. Statistical analysis was carried out by a one-tailed t test; * is for p ≤ 0.05; ** is for p ≤ 0.01; *** is for p ≤ 0.001.
