Synthetic genes were designed with, from 5′ to 3′, the SP6 bacteriophage promoter, a 5′ untranslated region (UTR), Kozak sequence, coding sequence of interest and 3′ UTR and transcribed into RNA. A 5′ Cap and poly (A) tail were enzymatically added to the purified single-stranded RNA to yield the mature mRNA.
