(A) i) P2SIY tumor bearing mice were treated with CD8 depleting antibodies one day prior to treatment with 12Gy radiation and intratumoral poly I:C. ii) Tumor growth and iii) animal survival were monitored following treatment. (B) i) Tumors were harvested 7 days following treatment with radiation plus poly I:C and CD8+ T cells were gated as Live CD45+ CD19 CD90.2+ CD4 CD8+. The number of intratumoral CD8+ T cells per mg of tumor tissue were quantified. ii) The expression of Ki67 and Granzyme B was assessed on all intratumoral CD8+ T cells and iii) within tumor antigen SIY+ CD8 T cells within P2SIY tumors. (C) i) P2SIY tumor bearing mice were treated daily with intraperitoneal FTY720 injections starting one day before treatment with radiation and poly I:C. ii) Tumor growth and iii) animal survival following radiation and poly I:C with FTY720 treatment. n = 4–7 animals/group.
