Small molecules bind to Hsp90 CTD and affect cancer cell viability. (a) ELISA. Microtiter wells were coated with the indicated increasing concentrations of small molecules and incubated with recombinant full-length or C-domain of Hsp90. Binding was determined using domain-specific antibodies to Hsp90 and quantified by absorbance at OD405. Data are the mean ±SEM of three independent experiments. (b) Inhibition of cell viability in H460 and PC3 cancer cell lines, and normal smooth muscle cells A10 and HUVEC cells, as evaluated by cell counting after a 24 h exposure to the selected small molecules.
