Inhibition of IL-1β-induced COX-2 expression in synovial fibroblasts depleted of p65 and p105. (A) Protein levels of total p65 (t-p65), total p105 (t-p105), and β-actin (internal standard) were detected by immunoblotting in cells transfected with p65, p105, or scramble siRNAs (control). (B) The relative levels of t-p65 and t-p105 as compared to the levels in the control samples. (C) Cells transfected with p65, p105, or scramble siRNAs were stimulated with IL-1β for 6 h. Depleting cells of p65 and p105 inhibited IL-1β-induced COX-2 expression. Results have been represented as mean ± SE from biological triplicates. *P < 0.05. Cell lysates (10 μg protein) were used for immunoblotting.
