Figure 6

IL-1β-induced activation of ERK1/2 signaling. (A) Western blotting for the levels of phosphorylated ERK1/2 (p-ERK1/2), total ERK1/2 (t-ERK1/2), phosphorylated JNK (p-JNK), total JNK (t-JNK), phosphorylated p38 MAPK (p-p38), and total p38 MAPK (t-p38) in cells treated with IL-1β (100 pM). Relative levels of p-ERK1/2, p-JNK, and p-p38 compared to the levels at 0 h (right panel). (B, C) Cell were pretreated with the ERK1/2 inhibitor FR180204 (50 µM; B) and MEK inhibitor U0126 (20 µM; C) for 1 h and stimulated with IL-1β for 15 min. MEK and ERK1/2 inhibitors significant reduced IL-1β-induced phosphorylation of ERK1/2. Relative levels of p-ERK1/2 as compared to those without IL-1β (right panel). Results have been represented as mean ± SE from biological triplicates. *P < 0.05. Cell lysates (10 μg protein) were used for immunoblotting. β-actin was used as an internal standard (A).
