Cytokine production by sorted DC subsets (putative cDC1, cDC2, and CD14⁺ DCs) following TLR stimulation. Cells were stimulated for 18 h with 10 μg/ml poly I:C, 1 μg/ml LPS, 10 μg/ml gardiquimod, or left unstimulated as controls. The supernatants were harvested and tested for IFN-α, IFN-γ, IL-4, IL-6, IL-12p40, IL-10, IL-1β, TNF-α, and IL-8 (CXCL8) by multiplex immunoassay. Production of IL-12p40, IL-10, IL-1β, TNF-α, IL-6, and IL-8 (CXCL8) was shown, while IFN-α, IFN-γ, or IL-4 were not shown as rarely detected. Alveolar macrophages (AMs) were used as a different cell-type reference. The values were displayed with logarithmic y-axis. Symbols in different colors represent different animals.
