Cellular retention of activity after interrupted incubation of [99mTc]Tc(CO)3-Ec1 (A) and [125I]I-PIB-Ec1 (B) with MDA-MB-468 cells. Cells were first incubated with the DARPin variants (10 nM) at 4 °C for 1 h and then the media were exchanged and the cells were incubated at 37 °C for 1, 4, or 24 h. A fraction of the supernatant at every time point was analyzed using NAP-5 size-exclusion columns and compared to the control when the radiolabeled DARPins were incubated in complete media for 24 h (C,D). Numbers in panels (C,D) show a percentage of activity associated with low-molecular-weight compounds at each time point. Data for retention are presented as the mean of three samples ± SD and data for NAP-5 analysis are presented as the mean of two samples ± SD. LMW = low molecular weight, HMW = high molecular weight.
