NDP-MSH binding properties of WT and mutant MC4Rs. HEK293T cells were transiently transfected with WT and mutant MC4Rs. Intact cell surface binding was measured by competitive inhibition of radiolabeled (iodine-125) NDP-MSH ([125I]-NDP–MSH) with different concentrations of unlabeled NDP–MSH. Results are expressed as % of WT maximal binding ± S.E.M. of duplicate measurements within one experiment. Curves are representative of three independent experiments.
