Effect of dinaciclib on its target genes. (A) Treatment of CHNG6 and established cell lines (HuCCT1 and KMCH) with varying doses of dinaciclib suppressed expression of CDK2, CDK5 and CDK9, the molecular targets of dinaciclib. (B) Dinaciclib treatment suppressed expression of anti-apoptotic genes BCL2 and BCL-XL and phosphorylation of RB in PDXC and two CCA cell lines. Quantification of western blot analysis was performed by Image J software and the numbers indicate the suppression in fold levels compared to vehicle treatment, denoted as 1.
