(A) Every residue of a receptor is mutated, and key pharmacological properties of the adrenaline-β2-adrenergic receptor-Gs signaling system are determined experimentally. (B, C) Active and inactive-state structures of receptors are analysed as networks of non-covalent contacts between residues to infer contacts that are specific for the active and/or inactive state. (D) These data are integrated using a data science approach to discover the molecular determinants and the underlying allosteric network governing efficacy and potency.
