(b) Huh7 cells were electroporated with replicon RNA and harvested into passive lysis buffer at the indicated time points. Luciferase activity was measured as previously described (Macdonald et al., 2003). Data from three independent experiments are shown; bars indicate SEM. (c) At 72 h p.t., samples were analysed by Western blotting with antibodies to NS3 (Aoubala et al., 2001), NS5A (Macdonald et al., 2003) or GAPDH. GND represents a corresponding polymerase-defective construct.
