(a) No accumulation of reduced wild-type EUGO is observed when the enzyme is aerobically incubated with vanillyl alcohol. The spectra before substrate addition and after one-minute incubation are in blue and red, respectively. (b) EUGO L152P (blue) is reduced by the addition of the substrate (red, one-minute incubation) and complete re-oxidation is observed only after 5 minutes (green). Protein and vanillyl-alcohol concentrations are 30 μM and 45 μM, respectively. Vanillin forms upon vanillyl alcohol oxidation (Fig. 2h).
