(a) Schematic representation of the enzymatic cascade used to assay FBPase activity. A coupled-enzyme assay including phosphoglucoisomerase (PGI) and glucose-6-phosphate dehydrogenase (G6PDH) was used to monitor FBPase activity by measuring the increase in absorbance due to NADPH production. (b) FBPase-related specific activity of the partially purified FBPase/SBPase in native (DTT 0 mM) or reducing (DTT 20 mM) conditions. (c) FBPase-related activity assayed with 0 or 32 mM of MgSO4 in the assay mix. Error bars are reported as standard deviations (n = 3).
