The solid blue curve (left y axis) in the main plot shows the steady-state relative enrichment of methylation, as predicted by our theoretical model. The inset shows the rescaled experimental data taken from mouse embryonic stem cells (i.e., H3K9me3 on day 5 divided by the average H3K9me3 on day 0) (16). The optimal parameters for our model are C_HP1,free = 1.37 × 10⁻⁵μM and an effective NFR of 1000 bp (fixing k_m/k_d = 0.042). The orange curve (right y axis) in the main plot shows the probability that a nucleosome forms a loop with the nucleation site as a function of genomic distance from the TSS.
