The four-amino-acid MA mutant signature can be introduced into subtype B to reduce PI susceptibility. Site-directed mutations were generated in the subtype B reference strain used in our assays. V128del was also added, as this deletion is present in HIV-1 CRF02_AG. Data are fold differences in IC50s of LPV in comparison to that for the assay reference strain, p8.9NSX.
