Scheme of MARS116-Bt binding to CatG. Cell surface CatG is drawn with the active-site cleft and the two β barrels. The hydroxyl group of S195 nucleophilically attacks the phosphorus atom of SucVPF and MARS116-Bt resulting in a covalent bond between S195 and the inhibitors. Anti-biotin-150Nd antibodies are added to the cell culture, forming the MARS116-Bt-anti-biotin-150Nd antibody complex, which can be analyzed by mass cytometry by time-of-flight (CyTOF).
