Loss of suppressor of Ty 16 (Spt16) impaired cell cycle progression and induced apoptosis in human lung cancer cells. A, A549 and NCI‐H1299 lung cancer cells were transfected with a control or two separate Spt16‐specific siRNA for 48 h. Flow cytometry (FACS) analyses with propidium iodide (PI) staining were performed to determine the effects of Spt16 on cell cycle progression. B, A549 and NCI‐H1299 lung cancer cells were transfected as in (A). EdU incorporation assays were performed. C, A549 and NCI‐H1299 lung cancer cells were transfected as in (A). Western blot analyses were performed with antibodies as indicated. This experiment was repeated three times independently. D, A549 and NCI‐H1299 lung cancer cells were transfected as in (A). Flow cytometry (FACS) analyses with Annexin‐V and 7‐AAD double staining were performed to determine the effects of Spt16 on cell apoptosis. E, A549 and NCI‐H1299 lung cancer cells were transfected as in (A). Western blot analyses were then performed with antibodies as indicated. This experiment was repeated three times independently.
