Effect of Atg5 siRNA on cell autophagy and apoptosis. A549 lung cancer cells were transfected with Atg5‐targeted siRNA (si ATG5) or control siRNA (si Control) for 48 h, then stimulated with peneciraistin C (Pe‐C, 3 μM) for 48 h. (a) Whole cell lysates were prepared and separated by SDS‐PAGE, then subjected to immunoblotting using indicated antibodies. (b) Cells were stained with FITC‐labeled annexin V and analyzed by flow cytometry. (c) Cells were treated with Pe‐C in the absence or presence of z‐VAD‐fmk (Z‐VAD). The cell death rate was measured by Trypan blue staining.
