Increased miR34a inhibits classical MHC-I expression in primary hippocampal neurons. (A) Representative images of transfection efficiency with Ctrl-GFP lentivirus or miR34a-GFP lentivirus infection in primary hippocampal neurons. Cells were infected with lentivirus at DIV 1 and observed at DIV 5 with a fluorescence microscope. Bars: 100 μm. (B) Cells were assessed for miRNA-34a expression at the mRNA levels after lentivirus infection. ***P < 0.001 vs. negative control. (C) Decreased H-2Db and H-2Kb in primary hippocampal neurons infected with miR34a-GFP lentivirus compared to those infected with Ctrl-GFP lentivirus for 4 days. The level of indicated mRNA was determined by RT-qPCR. *P < 0.05 vs. negative control. (D) Expression of the classical MHC-I protein in mouse primary hippocampal neurons infected with Ctrl-GFP lentivirus or miR34a-GFP lentivirus for 4 days as determined by western blotting. Densitometric data of the classical MHC-I protein expression using ImageJ are presented as the means ± SEM of three independent experiments. **P < 0.01 vs. negative control. All data are presented as the means ± SEM of three independent experiments.
