16S rRNA gene copy number quantification is influenced by the quality of the DNA mock. ZymoBIOMICS/humDNA mock at original DIN9 value and its six degradation levels corresponding to DIN7, DIN6, DIN4, DIN3, DIN2 and DIN1 were analysed for 16S copy quantification by ddPCR. The green line represents the total 16S copies ng−1 DNA, with this decreasing with the increase of the degradation level. The red line represents the 16S copy underestimation percentage, with this increasing with the DNA degradation level. 16S copies are reported as mean of at least three replicates for each sample±sd. 16S copy underestimation was calculated as a percentage compared to the highest quality (DIN 9) sample. **, P<0.01; ***, P<0,001.
