Glucose reduced expression of pentaglycine. S. aureus Newman was cultured in LB medium with or without 15 mM glucose for 24 h. Bacterial cells were collected by centrifugation and resuspended in PBS buffer, with lysozyme (A) or lysostaphin (B) added at a final concentration of 50 and 10 μg/mL, respectively. OD630 measurements were taken at timed intervals. (C) Lysostaphin resistance of S. aureus Newman in medium supplemented with 15 mM glucose was unaffected by the addition of 60 mM 2-DG. (D) S. aureus Newman was cultured in LB medium with or without 15 mM glucose for 6 h, and bacterial cells were collected for analysis of mRNA expression of femA, femB, and femX. ∗∗p < 0.01; ****p < 0.0001.
