Comparative detection of foot-and-mouth disease virus (FMDV) transcripts in draining popliteal lymph nodes (dPLNs) of inoculated mice. Comparison of transcript accumulation levels in dPLNs after inoculation of BALB/cJ mice. Animals (n = 3) were inoculated i.m. with 5 × 10⁸ PFU of Ad5[G], Ad5[P], or Ad5[P_VP2]_OP. Total RNA was extracted from dPLNs at 2 and 8 dpi, respectively, and a fragment of the FMDV 3C^pro open reading frame (ORF) and the GAPDH ORF were amplified by RT-qPCR. For each experimental group, the FMDV 3C^pro transcript level relative to the Ad5[P] group was calculated by using the 2^{− ΔΔCt} equation. Data correspond to the average and standard deviation from two independent experiments. *p < 0.01 (ANOVA).
