In vivo validation of Msx1-Fgf9/18-MAPK signaling in limb bud development. (A) qRT-PCR analysis to examine the expression profiles of Msx1, Fgf9, and Fgf18 in embryo limb buds during E9.5–14.5. Forelimb buds truncated from embryonic day 9.5–14.5 embryos were lysed. Total RNA was obtained and reverse-transcribed for qRT-PCR and RNA levels of Msx1, Fgf9 and Fgf18 were determined respectively. (B) Western blotting to detect the expression profiles of Msx1, Fgf9, Fgf18, PCNA, Ki67 and p-Erk1/2 in embryo limb buds during E9.5–14.5.
