(A) S. pombe strains were spotted onto YES or YEGES with or without 100 μg/mL Bz. Cells grown on YES for 1 day were washed three times. A culture with an OD600 of 2 was serially diluted from 10−1 to 10−5 (from left to right), spotted onto agar media, and cultured for 7 days. (B) For the pre-culture, PR110 yeast cells were cultivated in 55 mL of normal YES medium for 1 day, washed three times with pure water, and 100 μg/mL of benzoic acid (Bz) was added to YEGES media containing 2% (w/v) glycerol and 1% ethanol (w/v) instead of 3% glucose (w/v) with an initial cell density of ~1×107 cells/mL, and cells were cultivated for 3 days at 30°C. Gray bars show the CoQ10 content per 50 mL medium, and white bars show CoQ10 normalized against cell number. Five micrograms of CoQ6 was used as an internal standard for CoQ extraction.
