ARPE-19 cells were treated with HG (30 mM), HG (30 mM)+pcDNA3.1-TGF-β2, HG (30 mM)+pcDNA3.1 TGF-β2+miR-200a-3p mimic, HG (30 mM)+SRI (5 mM)+miR-200a-3p mimic. (A,B) The protein levels of TGF-β2, Smad2, and Smad3 were detected using Western blotting. (C) MTT assay was used to measure cell viability. (D,E) Flow cytometry was performed to examine cell apoptosis. (F) Western blotting was applied to detect the protein levels of Bax, caspase-3, Bcl-2, and VEGF.
