NaIO3-induced cell death is enhanced by antioxidants N-acetyl cysteine (NAC) and trolox. (A) Cells were pretreated with NAC (3 mM, 10 mM) 20 min prior to NaIO3 (30 mM) for 18 h. (B) Cells were pretreated with NAC (10 mM), trolox (500 μM) 20 min prior to NaIO3 stimulation for indicated time points (left panel) or 6 h (right panel). Cell viability was determined by Annexin V/PI staining using flow cytometry. (C,D) After pre-treatment with antioxidants followed by NaIO3 (30 mM) for 6 h, cytosolic H2O2 (C) and cytosolic O2− (D) were measured by using DCFDA and DHE, respectively. (E) After pretreatment with DPI (3 μM) 20 min prior to NaIO3 (30 mM) for indicated time points, cytosolic H2O2 was determined. (F) Cell viability was determined after cells were pretreated with DPI followed by NaIO3 (30 mM) for 18 h.
