NaIO₃-induced cell death is enhanced by antioxidants N-acetyl cysteine (NAC) and trolox. (A) Cells were pretreated with NAC (3 mM, 10 mM) 20 min prior to NaIO₃ (30 mM) for 18 h. (B) Cells were pretreated with NAC (10 mM), trolox (500 μM) 20 min prior to NaIO₃ stimulation for indicated time points (left panel) or 6 h (right panel). Cell viability was determined by Annexin V/PI staining using flow cytometry. (C,D) After pre-treatment with antioxidants followed by NaIO₃ (30 mM) for 6 h, cytosolic H₂O₂ (C) and cytosolic O₂⁻ (D) were measured by using DCFDA and DHE, respectively. (E) After pretreatment with DPI (3 μM) 20 min prior to NaIO₃ (30 mM) for indicated time points, cytosolic H₂O₂ was determined. (F) Cell viability was determined after cells were pretreated with DPI followed by NaIO₃ (30 mM) for 18 h.
