Conformational changes in the 14A mutant are very similar to those occurring in WT P-gp. (A) WT and 14A show comparable binding to UIC2 Fab in different steps of the ATP hydrolysis cycle. The histogram shows the quantification of bands corresponding to WT or 14A P-gp: UIC2-Fab complex (native-gels presented in SI Appendix, Fig. S8B).
