1. Telomere length and G‐overhang length analysis for HeLa cells transfected with pSpCas9(BB)‐2A-puro containing gRNA1 and gRNA2 sequences or the corresponding empty vector. The experiment was done in four replicates.
2. Western blot for POT1 and γH2AX upon POT1 deletion with gRNAs in HeLa cells.
3. C‐circle (CC) assay for HeLa cells upon POT1 removal. Bars represent data from four independent experiments ± SD. Significance was determined using one‐way ANOVA, and P‐values are indicated on the graph.
