Figure 5. FOXM1 and MYB pathways are upregulated in MC-CAR and 41BB-CAR T cells.

Figure 5

CAR T cells from 3 healthy donors were sorted into CD4+CAR+ and CD8+CAR+ subsets. RNA was extracted from sorted cells and sequenced on an Illumina NovaSeq platform. (A) Principal components analysis of the 3000 most variable genes. (B) Top 25 differentially expressed genes at baseline in CD4+ and CD8+ CAR T cells. (C) Western blot showing expression of MYB in unstimulated CAR T cells. GAPDH was used as a loading control. (D and E) Top 25 differentially expressed genes in each direction for unstimulated CD4+ (D) and CD8+ (E) 41BB- vs. MC-CAR T cells. (F) Genes differentially expressed between 41BB- and MC-CAR T cells were examined to identify upstream regulatory molecules and mechanisms that differentiate them using Ingenuity Pathway Analysis (IPA). The top 35 upstream regulators, ranked by P value, identified using IPA are shown. Enlarged versions of B and DF are provided in Supplemental Figures 18–21.
