Phenotype determination of different CD4+Treg subsets in tumor draining lymph nodes from patients with bladder cancer. To determine the frequency of different CD4+ subsets, CD4+ cells were first selected in the lymphocyte gate (A). Beside determining total frequency of CD4+FOXP3+ cells (B), CD4+CD25+ lymphocytes (C) which were FOXP3+ with low or no expression of CD127 were considered regulatory T cells (D); CD4+CD25+FOXP3+ cells that were CD127+ were also assessed (D). In some cases, this phenotype was dominant among CD4+CD25+ lymphocytes (E).
