Phenotype determination of different CD4⁺Treg subsets in tumor draining lymph nodes from patients with bladder cancer. To determine the frequency of different CD4⁺ subsets, CD4⁺ cells were first selected in the lymphocyte gate (A). Beside determining total frequency of CD4⁺FOXP3⁺ cells (B), CD4⁺CD25⁺ lymphocytes (C) which were FOXP3⁺ with low or no expression of CD127 were considered regulatory T cells (D); CD4⁺CD25⁺FOXP3⁺ cells that were CD127⁺ were also assessed (D). In some cases, this phenotype was dominant among CD4⁺CD25⁺ lymphocytes (E).
