Overview of the steps characterizing clustering-based methods. (A) Location is used to count mutations by protein domains or proximity in both coding and non-coding regions. (B) A statistical model is used to estimate the probability of observing the number of mutations by random chance. (C) Highly unlikely mutations counts are further annotated or analyzed. (D) Filtering by annotation scores finally determine report hotspots.
