(A) Western blot analysis indicates over-expression of MYC increases EZH2 expression in 957E/T-AR cells. (B) EZH2 inhibitor response curves indicate 957E/T-AR-MYC cells respond in a dose-dependent inhibition of cell growth following 3 days treatment. (C) Calculated Emax50 concentrations from (B) were used to treat cells for 3 days and show reduction of H3K27me3 levels in 957E/T-AR-MYC cells. (D) Emax50 concentrations were used to treat for 3 days prior to drug wash out to assess clonogenic potential in 957E/T-AR-MYC cells.
