Effect of vitronectin isoforms on HUVEC migration and tube formation. (A) To investigate an effect of vitronectin isoforms on endothelial migration, a scratch assay was performed with HUVECs cultivated in the presence of VTN_rs704:C, VTN_rs704:T, or control eluate (see Supplementary Fig. S5), and cell migration was defined as the percentage area closed after 14 hours of incubation. Data represent mean ± SD of six independent replicates, calibrated against the control. (B) HUVECs were cultivated in the presence of recombinant VTN_rs704:C, VTN_rs704:T, or control eluate. After 16 hours, the formation of tubular-like structures was followed microscopically (standard, 400 µm). The length of tubular-like structures was measured using ImageJ. Data represent mean ± SD of 11 independent replicates calibrated against the control.
