TGF-β regulates expression of fibrous tissue markers. (A) Primary sclerotome was treated with vehicle control or TGFβ1 for 2 h or 8 h. qPCR was used to determine the relative levels of Scx, Fmod and Adamtsl2 mRNA. (B, C) Sclerotome was treated with cycloheximide (CHX) for 30 min then treated either with or without TGFβ1 for 2 h (B) or 8 h (C). Relative levels of fibrous tissue markers were determined by qPCR. All mRNA levels were normalized to Hprt and then compared to vehicle control using REST software. (*) indicates significance, p < 0.05, n = 3.
