PP VII regulates macrophage transformation determined by STING. (a–j) RAW 264.7s were transfected with siSTING and siIRF3. 48 h later, cells were stimulated with IFN-γ or IL-6 in absence or presence of PP VII and analyzed by immunoblot (a,b,e). RAW 264.7s and THP-1s with or without PP VII were pretreated with indicated inhibitors and stimulated as in a (c,d,f). RAW 264.7s were treated with DMXAA for 3 h (g). siSTING-transfected RAW 264.7s were treated with DMXAA prior to indicated stimulation (h,i).
