Fgl2 regulates antibody responses in vivo. (a) Serum antibody isotypes in 20-week-old wild-type and Fgl2−/− mice were detected by bead-based mouse immunoglobulin isotyping kit and ELISA (for IgE). The mouse immunoglobulin isotyping kit provides no standard curve assessment so the results should be considered more as qualitative. (b-c) Peyer’s Patches of 20-month-old wild-type and Fgl2−/− mice were immunophenotyped by flow cytometry for IgA+ B cells, GC B cells, TFH cells, TFR cells and total Treg cells. Free fecal IgA was measured by ELISA. (d) Wild-type and Fgl2−/− mice were immunized with NP-OVA (s.c.) in CFA for 21 days and NP-specific isotypes were detected by ELISA. The results were pooled from 3 independent experiments. Differences between two groups were compared using two-tailed unpaired Welch’s T tests (n.s; * p <0.05, ** p<0.01 and *** p<0.001). All plots show the means ± SD.
