Single-Cell Dynamics of IFN-β and ISG15 Upregulation in Response to HCMV or SeV Particles

(A–E) IFN-β-GFP THFs were pretreated with B18R or media alone and mock treated, treated with IFNα (1000 U/ml), infected with HCMV-UV (1,344 particles/cell), or infected with SeV-UV (200 particles/cell). Cells were fixed 16 hr later and levels of GFP and ISG15 expression measured by flow cytometry (A–D). Cells were categorized as GFP^low/high and ISG15^low/high based on measurements of mock treated cells and mean proportion and standard error from 3 biological replicates graphed. Similarly, treated IFN-β-GFP THFs were visualized by fluorescence microscopy, and representative images from 3 biological replicates are shown (E). Scale bars of 400 μm are shown.
