Figure 3

UVA radiation-intensified lomefloxacin induces ROS production in amelanotic C32 (A) and melanotic COLO829 (B) melanoma cells. The cells were pre-treated with lomefloxacin at concentrations of 0.1, 0.5, and 1.0 mM alone for 24 h or exposed to the drug and UVA irradiation (1.3 J/cm2). The data are expressed as percentages of the controls normalised to a number of living cells. Mean values ± SEM from three independent experiments (n = 9) performed in triplicate are presented. * p < 0.05, ** p < 0.005 vs. control samples. Co-treatment of C32 (C) and COLO829 (D) melanoma cells with lomefloxacin and UVA irradiation augments cellular GSH depletion. The cells were pre-treated with lomefloxacin at concentrations of 0.1, 0.5, and 1.0 mM alone for 24 h or exposed to the drug and UVA irradiation (1.3 J/cm2). Mean values ± SEM from three independent experiments (n = 9) performed in triplicate are presented. ** p < 0.005 vs. control samples. Representative histograms are available in the Supplementary Materials S1.
