Withaferin A (WA) treatment modulated expression of genes associated with cellular senescence. A, Expression of retinoblastoma transcriptional corepressor like 2 (RBL2), nibrin (NBN), nuclear factor of activated T cells 4 (NFATC4), protein phosphatase 1 catalytic subunit gamma (PPP1CC), and ZFP36 ring finger protein like 2 (ZFP36L2) genes in four cell lines after 16-hour treatment with dimethyl sulfoxide (DMSO, control) or 2 μM WA. Results shown are mean ± SD (n=3) and statistical analysis was done by unpaired Student’s t-test (*, P < 0.05). B, Quantitative real-time reverse transcription polymerase chain reaction analysis of above-mentioned genes in breast cancer cell lines treated with DMSO (control) or 2 μM WA for 8 and 16 hours. Combined results from two independent experiments are shown as mean ± SD (n=6). Statistical analysis was done by unpaired Student’s t-test (*, P < 0.05).
