Nrf2 knockdown abolished the protective effects of eugenol on H2O2-induced damage. (A) The HEK-293 cells transfected with plasmids expressing shNRF2 or shControl were treated with eugenol (0, 25, 50 or 100 µg/ml) and H2O2 (0, 50, 100 or 250 µM) for 24 h, following which MTT assays were performed to measure cell viability. (B) An identical experiment was also performed in NIH-3T3 cells but with 100 µg/ml eugenol. The IC50 values were calculated according to the obtained MTT assay data. tBHQ (100 µg/ml) was used as positive control. Experiments were performed in triplicate. Data were presented as mean ± SD. *P<0.05 vs. shNRF2. Nrf2, nuclear factor erythroid 2-related factor 2; tBHQ, tert-butylhydroquinone; sh, short hairpin RNA.
