A) Succinylated EchA19wt (5 μM) was incubated with 2 μM Rv1151c in the presence of 1 mM NAD+ in a solution containing 25 mM Tris-HCl (pH 7.5), 137 mM NaCl, 2.7 mM KCl, 1 mM MgCl2, and 0.1% BSA. Desuccinylase activity catalyzed by Rv1151c was monitored as a function of time over 2 h, and remaining succinylated EchA19 was detected with a pan anti-succinyllysine antibody. B) Densitometry analysis of blots from three replicates of experiment shown in (A). Intensity was scaled to 100% for succinylated EchA19wt. Intensity was scaled to 100% for succinylated EchA19wt. Errors shown are the standard error of measurement.
