cc-del6 Orientation in Conjunction with pro-β Motif Favors the Production of SDS-Resistant Oligomers by γ-Secretase Processing
(A) Analysis of the profile of cc-del(X) constructs in denaturing conditions. Cells lysates were analyzed by Western blotting and revealed with APP Cter antibody (left) or HA antibody (right).
(B) Western blot of MEF APP+/+, MEF double knockout for presenilin 1 and 2 (dPSEN−/−). Detection was performed using the HA-tag antibody.
(C) Analysis of extracellular media. Immunoprecipitation was performed on the supernatant. CHO cells were expressing HA-C99 and cc-del(X) constructs, analyzed by Western blotting and revealed with HA antibody (top). At high exposure (bottom), monomeric Aβ-like (Aβ∗) was detected.
(D) Neuronal lysates were analyzed 4 days after infection by Western blotting and revealed with HA antibody.
(E) Top: Representation of HA-C42/43 and cc-Aβn−42/43 constructs. Bottom: Analysis of the profile of truncated forms (C42/C43) of cc-del(X) constructs and Aβ-like peptides (top) in denaturing conditions. Cell lysates were analyzed by Western blotting and revealed with HA antibody (bottom).
