Screening of the EEBL library with the human colon organoids in a 384-well plate format. Human colon APC−/−;KRASG12D organoids cultured in 384-well plates were treated with library compounds (0.1 µl, final at 4.6 µM). The plates were incubated for 3 days and subjected to CellTiter Blue cell viability assay. The effect of compounds on the growth of KRASG12D organoids was calculated as % Inhibition based on each plate and analyzed using Cambridge BioAssay software.
