SP enhances vitamin D–dependent inhibition of NF‐κB activity. RAW 264.7 reporter cells were activated with 100 ng/mL LPS and 20 ng/mL IFN‐γ. One hour later, cells were treated with VD (calcitriol) in the absence/presence of SP as indicated. Absorbance was read after 24‐h incubation. (A) NF‐κB activity reduced by 500 nM VD (calcitriol) treatment. (B) Combined treatment of the cells with SP and VD (calcitriol) further decreased NF‐κB activity. SP was used at 10 μM concentration, and Et‐OH served as a vehicle. The average ± SEM; n = 4. VD, calcitriol; SP, spironolactone.
