Role of IL-2 receptor in activating signal transmission. (a) Lymphocytes expressing IL-2Rα (CD25), IL-2Rβ (CD122) and IL-2Rγ (CD132) subunits were removed from PBMCs by magnetic separation, and the remaining cells were incubated with Tag7 for 6 days and tested for cytotoxicity against K562 cells. (b) JAK–STAT pathway is involved in PBMC activation by Tag7. PBMCs were incubated with Tag7 for 6 days in the presence of specific inhibitors of JAK1, JAK3, STAT3 and STAT5 (added 1 h before treatment and additionally added on days 3 and 5 in the case of STAT3 and STAT5) and tested for cytotoxicity against K562 cells. PBMCs after 6 days incubation with Tag7 tested for control cytotoxicity. Data are presented as the mean ± SD of 3 independent experiments. Differences from the control in all cases are significant at * p < 0.05 (2-way ANOVA).
