Effect of Jakyakgamcho-Tang (JGTW) and 30% EtOH Jakyakgamcho-Tang (JGTE) on viability, nitric oxide (NO), prostaglandin (PG) E2 production, and inducible nitric oxide/cyclooxygenase-2 (iNOS/COX-2) expression of lipopolysaccharide (LPS)-induced RAW264.7 macrophages. RAW264.7 cells were pre-treated with JGTW or JGTE (50, 100, and 200 µg/mL) for 2 h, and then stimulated with LPS (0.5 µg/mL) for 24 h. (A) Cell viability was measured using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. (B) NO production was measured using a Griess reagent assay. L-NG-monomethyl arginine (L-NMMA) was used as a positive control. (C) PGE2 was measured using ELISA. The levels of (D) iNOS and (E) COX-2 were measured by real-time PCR.
