Effect of hydrogen sulfide donor on diabetes-induced alterations in H2S and its enzymatic machinery. Retina from diabetic mice, with or without homocysteine and GY supplementation, were analyzed for (a) H2S and (b) homocysteine levels using their respective colorimetric methods. (c) CBS activity was quantified using cysteine and homocysteine as substrates, and (d) CSE activity using L-cysteine as a substrate and pyridoxal-5′-phosphate as a cofactor. Each measurement was made in duplicate in 5 to 7 mice/group, and the values are represented as mean ± SD. Norm, normal; Diab and Diab/GY, diabetic mice without or with GY respectively; Diab/H and Diab/H + GY, homocysteine supplemented diabetic mice, without or with GY. *P < 0.05 versus normal, #P < 0.05 versus diabetes, and ^P < 0.05 versus diabetic mice receiving homocysteine.
