Generation of inducible endothelial-specific WWOX knockout (KO) mice. WWOX flox/flox mice were obtained from the laboratory of Dr. C. Marcelo Aldaz, M.D., Ph.D., of MD Anderson Cancer Center at the University of Texas. These were bred with mice containing a tamoxifen-inducible cre deleter under the control of the CDH5 promoter to obtain an inducible endothelial-specific WWOX KO. (A) Genotyping was performed as described in Ludes-Meyers and colleagues (24). (B) Lung endothelial cells were isolated from cell suspensions obtained from WWOX flox and KO mice using FACS to segregate CD45 CD31+ cells. These were grown in culture and analyzed for WWOX expression by RT-PCR (C) and Western blotting (D). Results are shown as means ± SD in n = 3 independent experiments. *P < 0.05, compared with control by Student’s t test. f/f = flox/flox; WX = WWOX; WT = wild-type.
